Knock-in pig models of Huntington's disease-causing genes can more realistically reproduce neurodegeneration in HD patients. The evidence of  lesions in the HD KI pig model provides a basis and effective treatment strategy for the development of animal models to simulate selective neurodegeneration in other key neurodegenerative diseases, such as AD and PD.

The hSOD1 transgenic pigs showed disease characteristics typical of human ALS and unique nuclear inclusions that have not been seen in other animal models of ALS. Transgenic pigs would provide a useful model for investigating the pathogenesis of ALS and identifying potential therapeutic targets.

Mutant TDP-43 was found to be distributed in the cytoplasm of brain tissue of transgenic pigs, and this phenomenon was also seen in the brains of patients. By studying the interaction of TDP-43 with PSF and its associated RNA splicing function, we provide new evidence for the species-dependent cytoplasmic distribution of mutant TDP-43 and gained new insights into how cytoplasmic accumulation of TDP-43 mutants can lead to abnormal gene expression.

NLRP3 R259W homozygous pigs had a typical delayed hypersensitivity reaction (DTH), and the mutant pigs showed a more intense inflammatory response to 5% DNCB solution (24h after application), accompanied by skin shrinkage and redness, and a longer response time.

Shank3 gene-edited dogs exhibited core symptoms of autism such as social deficits (reduced social time, increased social pressure) and stereotyped repetitive behaviors during conspecific (the three-chamber test) and dog–human interactions.

The Shank3 mutant dogs recapitulate key phenotypes associated with ASD, which make them an ideal animal model for autism research.
